Abstract
Tuberculosis is a serious illness where its cure lies on early diagnosis. One effective diagnostic tool is fluorescence digital microscopy which is a commonly used technique consists of analyzing sputum images for detecting bacilli. The analysis of sputum is time consuming and requires highly trained personnel to avoid high errors. Image-processing techniques provide a good tool for improving the manual screening of samples. In this thesis, a new autofocus algorithm and a new bacilli detection technique is developed and presented with the aim to attain a high specificity rate and reduce the time consumed to analyze such sputum samples. This technique is based on segmentation used with some shape features together with a simple thresholding operation on the RGB image after converting it into gray scale image. Some feature descriptors have been extracted from bacilli shape using reference database image. A sobel edge detection technique was applied for image analysis purposes and the accuracy results were evaluated using a standard SURF (speeded up robust feature) analysis procedure.
